Statement:
My background is in structural and functional studies of mitochondrial and bacterial respiratory complexes, especially cyt bc1 complex. The bc1 complex of Mycobacteria is highly divergent from the mitochondrial one and forms a supercomplex with the cyt aa3 oxidase. This enzyme has not been well characterized. Recently a promising class of antitubercular drugs was shown to target the bc1 complex. In collaboration with scientists at the Pasteur Institute, Korea, I have started a project to isolate, characterize, and solve the crystal structure of the bc1 complex of M. tuberculosis, (probably as the bcc-aa3 supercomplex). If successful this will delineate how these drugs bind in the bc1 complex and be useful for design of improved members of this promising new class of drugs.
